Supplemental Material:

Videos. Transmigration of cells across an endothelial layer. Time-lapse video microscopy, as described in "Materials and methods," was used to visualize migration of selected CD34+ and CD8+ cells across a bone marrow endothelial layer[en]derived cell line. Cells were either visualized alone or in combination by labeling CD8+ cells with CFDA-SE.

Video 1 (MOV file, 1.4MB): CD34+ alone.

Video 2 (MOV file, 2.7MB): CD8+ alone.

Video 3 (MOV file, 2.9MB): CD34+ and CD8+.

JPEG ImageFigure S1. Flow cytometry plots of CD34+ cells which have homed to the bone marrow. Selected CD34+ cells were labeled with CFDA-SE, incubated alone, with CD8+ cells, or with CD4+ cells, and then injected into sublethally irradiated mice. After 9 hours the bone marrow was analyzed for the presence of injected CD34+ cells by flow cytometry. One million events were collected on the flow cytometer from each experiment; only a proportion (5%) are displayed.

JPEG ImageFigure S2. Analysis of the level of engraftment by flow cytometry. Cells were mixed together then injected into sublethally irradiated mice. Six to eight weeks following injection, the mice were euthanized and the level of engraftment was measured by flow cytometry for the human CD45 marker.
