Recombinant P-selectin glycoprotein ligand-1 directly inhibits leukocyte rolling by all 3 selectins in vivo: complete inhibition of rolling is not required for anti-inflammatory effect

Movies 1-3 show leukocyte rolling in surgically stimulated venules of a mouse cremaster. Movie 1 (recorded 30 minutes after surgical preparation of tissue) shows control rolling. Movie 2 (the same vessels 2 minutes 30 seconds later) shows residual rolling after intravenous treatment with 100 mg/kg rPSGL-Ig. Movie 3 shows almost complete absence of rolling in the same vessels after treatment with P-selectin–blocking antibody. Movie 4 shows leukocytes forming new attachments to postcapillary venular endothelium soon after leaving capillaries.
