This document contains 3 figures that provide additional data on the mechanism of action of the anti-FLT3 antibody IMC-EB10. Contents include detailed descriptions of the methods and results of these experiments. Figure S1 shows that IMC-EB10 does not induce apoptosis in EOL-1 and BaF3-ITD leukemia cells. Figure S2 shows that IMC-EB10 does not affect the expression of the FIP1L1-PDGFRA fusion gene in EOL-1 cells. Figure S3 shows that incubation with IMC-EB10 antibody causes the internalization of FLT3 receptor on BaF3-ITD cells, suggesting that antibody-mediated internalization and the resulting down-modulation of surface FLT3 receptor may contribute to the anti–FLT3-ITD activity observed in the BaF3-ITD cell line.
