Figure S1. Distribution of PTPN11 and RAS mutations and TEL-AML1 fusion gene by DNA content and age at diagnosis in children with B-cell precursor ALL (JPEG file, 65 KB): (A) DNA content. PTPN11 and NRAS/KRAS2 gene mutations show a significant higher prevalence among children with hyperdiploid DNA content compared with those with DNA Index lower than or equal to 1 (PTPN11: 14% vs 4%, Pearson
2 = 9.97, P = .002; RAS genes: 35% vs 19%, Pearson 2 = 7.84, P = .005), whereas TEL-AML1 gene rearrangements are almost exclusively associated with diploid DNA content (3% vs 33%, Pearson 2 = 29.78, P < .001). White bars indicate DNA Index less than or equal to 1; solid bars indicate DNA Index greater than 1. (B) Age at diagnosis. Prevalence of PTPN11 mutations is significantly higher among children with age at diagnosis greater than or equal to 15 years compared with the other classes (P = .022, Fisher exact test), whereas prevalence of the TEL-AML1 fusion gene is restricted to children younger than 15 years, with highest recurrence among children between 5 and 10 years old (P = .005, Fisher exact test). No association between age at diagnosis and prevalence of RAS gene mutations is observed (P = 0.819, Fisher exact test). Age at diagnosis (in years) is: white bars, < 5; light gray bars, ≥ 5 and < 10; dark gray bars, ≥ 10 and < 15; and solid bars, ≥ 15.
