Both biopsies from each animal were cut (slice thickness 5 µm, left panel) into serial slices. Slices spanning the region of TNF-
injection were digitalized in 45- to 70-µm intervals using a microscope equipped with a digital camera. Image analysis was performed with digital image analysis software (KS 300; Carl Zeiss, Jena, Germany).
The outer perimeter of the inflamed area from each tissue section was specified manually in a blinded fashion. Blood vessels were marked and manually exluded; fat vacuoles of adipocytes were not covered by the image analysis program. The region specified in this way was overlaid with a black mask, which is illustrated by the right upper and lower panels, respectively. The area of the mask was determined for several slices. Peak values from 5 to 8 consecutive slices that covered a layer thickness of 300 µm of the biopsy in total were averaged.
