Clones were generated by single-cell sorting from PBLs stained with Vδ2, CD45RA, CD16, CD62L, and CD27. Cells were cultured as previously described.1 Expression levels of chemokine receptors by Vδ2 T-cell clones following 24 hours of stimulation with 10 nM BrHPP and 50 U/ml IL-2 were evaluated with flow cytometry.

The Vδ2high and Vδ2low phenotypes are stable in culture and correspond to TEMh and TEMRA subsets, respectively. Vδ2 T-cell clones were established following single-cell sorting based on TCR Vδ2 expression levels. The phenotypes were monitored in culture, and clones were stimulated with 20 nM BrHPP at 37°C at three time points.
