-MEM is better for hematopoietic differentiation. R1 ES cells were differentiated in -MEM or in IMDM. Day-3 EBs were subjected to FACS analyses for Flk-1 expression. (B) Mitomycin Ctreated OP9 cells support better ES differentiation. R1 ES cells were differentiated on mitomycin Ctreated or nontreated OP9 cells. On day 3, cells were harvested and subjected to FACS analyses for Flk-1 expression. OP9 cells and day-3 EBs were used as a gating control. ES cells differentiated on mitomycin Ctreated OP9 cells had a higher percentage of differentiated cells (50% vs 30%) and a higher cell number (average, 2- to 3-fold more; not shown). (C) OP9 differentiation is less sensitive to different lots of fetal calf serum. R1 ES cells were differentiated in preselected or nonselected fetal calf serum. Three days later, Flk-1 cells generated in EBs and on OP9 were analyzed by FACS. Flk-1+ cells generated greatly reduced in EBs when nonselected serum was used, while OP9 differentiation was less sensitive to serum. Another 2 lots of nonselected serum showed similar results (not shown).
