The intensity of Tcl1 staining is shown as a solid line and control antibody staining as a fine line in the figure. The results shown are representative of the three independent experiments. (A) Bone marrow cells were stained with anti-B220, anti-CD43, anti-IgM, or anti-IgD and then stained with 5A4 mAb, anti-Tcl1, intracellularly after fixation (2 % PFA) and permeabilization (0.1 % saponin). (B) Thymocytes were stained with anti-CD4 and anti-CD8, and then cytoplasmic staining with 5A4 mAb was followed. (C) To show the expression of Tcl1 in the double-negative thymocyte subpopulations, thymocytes were stained with anti-CD4/CD8, anti-CD44, and anti-CD25. After intracellular staining with 5A4 mAb and an isotype-matched control antibody, CD4–CD8– populations were gated and divided into four subpopulations based on CD44 and CD25 surface marker antigens.
