The GFP+ and GFP- (A) pDC subsets described in Figure 6 were sorted, and 1,000 cells were cultured in Iscove's MDM-based methylcellulose for 8 days. Colonies were then classified as BFU-E, CFU-GM and CFU-GEMM. (B) pDC subsets were sorted and 5,000 cells were cultured in stromal cell- and FCS-free culture medium with growth factors. Cells were harvested and stained with antibodies to CD19 and DX-5. BM Lin-c-Kit+ cells were cultured as positive control. (C) pDC subsets were sorted and cultured for two weeks with SCF and IL-7 in a fetal thymus organ culture system. Embryonic day 12 fetal thymus was used as a positive control.

pDC from spleen and BM of homozygous RAG1/GFP knock-in mice were resolved into GFP+ and GFP- populations (top histograms). Dotted lines: pDC from WT C57BL/6 mice. cDNA samples from sorted pDC1 and pDC2 of heterozygous mice were evaluated for RAG1 expression by RT-PCR (bottom panel) and compared to BM pro-B cells.
