Differential regulation of virus-specific T-cell effector functions following activation by peptide or innate cytokines

Differential regulation of cytokine production levels (PDF, 99 KB) - Spleen cells isolated from mice at 8 days (Day 8) or 145 days (Immune) after LCMV infection were activated directly ex vivo with either peptide alone or the combination of peptide plus IL-12 and IL-18. BFA was added for the final hour of stimulation, and cytokine production by CD8+ T cells was detected by intracellular staining. Each cytokine was analyzed at the peak time of production. (A) IFN production after 8 hours of stimulation. (B) TNF after 2 hours of activation. (C) CD40L after 3 hours of activation. (D) IL-2 after 3 hours of stimulation. Graphs depict MFI values for each cytokine and are the mean ± SD of 4 mice in each group. Statistical comparisons were made using a paired Student t test.
