Haploinsufficiency of GATA-2 perturbs adult hematopoietic stem-cell homeostasis

  • Figure S1. Reduced numbers of hematopoietic progenitor cells within the granulocyte-macrophage (GM) compartment of GATA-2+/- bone marrow (PDF, 29 KB) - Bone-marrow–nucleated cells from each genotype were assessed for total progenitor-cell number (CFCs), granulocyte-macrophages (CFC-GMs) and erythroid burst-forming unit progenitors (BFU-Es) as detailed in "Materials and Methods." Cells were plated at a density of 104 per mL per well in a six-well plate in CFC and CFC-GM assays. In the BFU-E assay, cells were plated at a density of 2 × 105 per mL per well in a six-well plate. Four replicates were used per genotype in each experiment. CFCs and CFC-GMs were scored on day 10 and BFU-Es were scored on day 7. The absolute number of total CFCs (A) (P=0.022; n=7), CFC-GMs (B) (P=0.009; n=5) and BFU-Es (C) (P=0.658; n=4) are shown. Statistical analysis was performed using the Student t-test.

GATA-2 real-time RT-PCR has a similar efficiency to that of GAPDH RT-PCR (PDF, 54 KB) - A five-fold dilution series of cDNA from total bone marrow of a wild-type mouse was prepared, and a real-time PCR for GATA-2 (A) and GAPDH (B) was performed. Representative amplification plots and standard curves demonstrate similar PCR efficiency for GATA-2 and GAPDH (97.2% and 99.5% respectively).
