A subset of natural killer cells achieves self-tolerance without expressing inhibitory receptors specific for self-MHC molecules

  • Figure S1. CI/NKG2- NK cells respond poorly when stimulated with Kb–/–Db–/– lymphoblasts (JPG, 46 KB) -

    Splenocytes from poly(I:C)-treated 2m–/– or B6 mice were stimulated for 5 hours in the presence of brefeldin A with Con A–induced lymphoblasts from the designated mice before staining the cells with mAbs specific for cell surface markers and intracellular IFN-. Results are depicted as the percentage ± SEM (n=3 mice) of IFN-+ cells in each subset (CI/NKG2+ or CI/NKG2–). Neither 2m–/– nor B6 CI/NKG2– NK cells responded significantly to Kb–/–Db–/– Con A blasts.

  • Figure S2. Bone marrow rejection assay with limiting doses of bone marrow (JPG, 53 KB) -

    Recipient mice (2m–/– or B6) were treated with mAbs to deplete the indicated NK cell subset, followed by lethal irradiation. At day 0, a mixture of 5 × 106 each (left panel) or 2.5 × 106 each (right panel) CFSE-labeled bone marrow cells from 2m–/– Ly5.1+ and B6 (Ly5.2) mice was injected intravenously. After 3 days, splenocytes were stained with PE-conjugated anti-Ly5.1 Ab. The results are represented as graft acceptance, calculated as the ratio of 2m–/– (Ly5.1+) bone marrow cells to B6 (Ly5.1–) bone marrow cells among gated CFSE-positive cells. The data represent means ± standard errors of the means of the ratios (n = 3). The B6 cells serve as a nonrejected internal reference population. The graft acceptance ratios in control groups vary from experiment to experiment, probably due to counting errors.
