Files in this Data Supplement:
- Video S1: Dynamics of tetraspanin CD9 during T lymphoblast transendothelial migration (MOV file, 2,567 KB)
-
Timelapse video sequence of lymphoblast transmigration through CD9-GFPtransfected TNF-
activated HUVEC monolayers during a total observation time of 30 minutes. The maximum projection of the informative sections from the GFP confocal image stack is merged with DIC image. White arrows mark the position of apically-adhered lymphocytes. Black arrows point to transmigrated lymphocytes interacting with the ventral surface of endothelium. CD9-GFP was more clearly relocalized to the contact site with T lymphoblasts at the endothelial apical surface, although some clustering occurred around transmigrated lymphoblasts locomoting underneath the endothelium.
- Video S2: Dynamics of tetraspanin CD151 during T lymphoblast transendothelial migration (MOV file, 1,754 KB)
-
Time-lapse video sequence of lymphoblast transmigration through CD151-GFPtransfected TNF-
activated HUVEC monolayers during a total observation time of 20 minutes. The maximum projection of the informative sections from the GFP confocal image stack is merged with DIC image. White arrows mark the position of apical adhered lymphocytes. Black arrows point to transmigrated lymphocytes interacting with the ventral surface of endothelium. CD151-GFP was strongly concentrated at the contact with lymphoblasts throughout all the transmigration process so that relocalization around lymphoblasts was clearly detected at both the apical and basal surface of the endothelial cell.
