    (A) Purified B cells or sorted T1 B cells were cultured for 1, 3 or 6 days with various concentrations of BAFF and the percentage cell survival was determined by flow cytometry using forward/side scatter characteristics and 7-AAD exclusion. (B) Human spleen B cells were enriched by negative selection and cultured with various stimuli for 3, 6 and 9 days and cell survival was determined. (C) Ig production was determined from splenic B cell supernatants after 6 days of culture by ELISA. SAC is Staphlococcus aureus Cowan I strain.

    The frequency of V, D and J gene segments was determined for productive V-D-J rearrangements from the combined T1 B cell population isolated from bone marrow (n=26), and peripheral blood from a normal donor (n=23) and an SLE patient (n=21).
