Donor origin of multipotent adult progenitor cells in radiation chimeras

    Mononuclear BM cells, isolated 1 month after sex-mismatched transplantation of female GFP-positive donor BM cells into a male host, were cultured in MAPC media. MAPC colonies from passage 3 were sorted as GFP-positive or GFP-negative. FISH analysis was used to detect the Y-chromosome with an FITC-conjugated probe (A, C, and E); a Dapi counterstain (B, D, and F) was used to visualize all genetic material. (A-B) All GFP-negative host cells contained the Y-chromosome. (C-F) None of the GFP-positive donor cells contained Y-chromosomes. Arrow indicates chromosomal spread 40, XY.
