(A) Inhibition of Kit-dependent cell proliferation by AP23848. Juxtamembrane and activation loop mutant cell lines were grown in graded concentrations of AP23848 for 48 hours, and viable cells were measured by MTT assay. Results are expressed as percent of control. Graphs represent the average of three independent experiments. (B) Effect of AP23848 on hematopoietic colony formation. Normal bone marrow mononuclear cells were incubated in semisolid media in the presence of SCF, GM-CSF, and IL-3, with or without erythropoietin, with escalating AP23848 concentrations. Granulocyte/macrophage (CFU-GM) and erythroblast (BFU-E) colony numbers are reported for one marrow sample, as a percentage of the untreated culture. Each data point represents the average of duplicate cultures.
