(A) The progeny of sorted CD34+/Lin– cells was analyzed after 3 weeks of culture in lymphomyeloid conditions. Results are expressed as the percentage of CD19+ B cells and CD15+ myeloid cells detected in 2 independent experiments. (B) Sorted CD34+/Lin– cells were cultured in methylcellulose for the detection of human CFCs. Results are expressed in absolute numbers of CFCs generated for 2000 plated cells. (C) CD34+/Lin– cells were cultured at limiting dilutions in LTC conditions. Frequencies of LTC-IC were calculated according to Poisson statistics and are indicated for GFP and TAL1 transgenes. Arrows indicate the cell number giving 37% negative wells (corresponding to the solid horizontal line) and thus indicate the LTC-IC frequency for each condition in 2 independent experiments (dashed and solid lines). Typical colonies generated from LTC-IC are shown (magnification, × 400). (D) mRNA levels of Bmi-1, HOXB4, and GATA-2 in CD34+/Lin– cells were measured by semi-quantitative RT-PCR analysis; representative of 2 independent experiments.
