The blue staining represents expression of the -galactosidase reporter gene. Figure S2. Reporter gene expression in hematopoietic cells of ROSA26R/TCre mice (JPG. 114 KB) - Cells from bone marrow, thymus, and spleen were loaded with fluorescein di-D-galactopyranoside, a reporter for -galactosidase activity, and then stained with PE-conjugated monoclonal antibodies to the B cell marker B220, the myeloid cell marker Mac-1, or the T cell marker Thy1.2. The cells were then analyzed by flow cytometry. Gates were set to demarcate cells that were positive or negative for the indicated marker. The percentage of cells expressing a specific combination of markers is shown in each gate.
