    (A) Western blot detection of Scurfin in lysates of A293T cells transfected with PG, a murine leukemia virus–based eGFP-expression vector, or PG-mFoxP3, expressing the murine FoxP3 cDNA under control of the viral LTR. Detection with anti–Scurfin antiserum, compared with preimmune serum. (B) Detection of Scurfin in A293T cells transfected with PG or PG-mFoxP3, by intracellular staining and FACS, using affinity purified antibody. (C) Total thymocytes from WT and Scurfy (Sf–/–) mice were stained for CD4, CD8, CD25 and intracellular Scurfin (mFoxP3) and were analyzed by flow cytometry for the percentage of CD4+CD8CD25+ cells expressing intracellular Scurfin.
