  • Figure S1. Evaluation of PCR sensitivity (PDF, 16.9 KB) - In experimental conditions characterized by the lowest amount of DNA allowed for the method, a percentage of contaminating plasma cells of at least 1% is necessary to give PCR positive results. The sensitivity of the method is evaluated by analyzing negative samples with decreasing percentages of clonal plasma cell (15%, 1%, 0.1%, 0.01%). Positive PCR results are obtained only for a percentage of contaminating plasma cells higher than 1%, whereas a weakly positive band is visible at 1% contamination. Note that in all immunological analyses the maximum percentage of positivity for plasma cell associated antigens (CD38 and CD138) is 0.5%, while the mean value of clonal CECs with 13q14 deletion is 18% (PCs: plasma cells).
