Cytomegalovirus MCK-2 controls mobilization and recruitment of myeloid progenitor cells to facilitate dissemination
Blood Noda et al. 107: 30

Supplemental materials for: Noda et al

Files in this Data Supplement:

  • Figure S1. FCM analysis of Mac-1+Gr-1int cell patterns in draining lymph nodes and spleen following MCMV infection (JPG, 106 KB) -

Leukocytes prepared from popliteal lymph node (Panel A) and spleen (Panel B) of groups of five infected BALB/c mice were subjected to FCM analysis described in Figure 5. Mac-1+Gr-1int cell populations are indicated by the boxed areas.

  • Figure S2. MCMV replication and dissemination in CCR2-/- mice (JPG, 57.9 KB) -

    A. Viremia levels determined by infectious centers assay on groups of three mice at day 5 post inoculation (i.p. route) with 105 PFU (SG propagated virus stock) of repaired virus RQ461 (filled triangle) or mck-mutant RM461 (open triangle) in C57BL/6 and CCR2-/- mice. Mean (line) and standard deviation are indicated along with data from individual mice. B. Titers of virus in spleen, lungs and liver from the RQ461 infected mice used in A.

  • Figure S3. MCMV replication and dissemination in MCP-1-/- mice (JPG, 61.2 KB) -

    A. Cell-associated viremia measured by ICA on groups of three BALB/c or MCP-1-/- mice at day 5 post inoculation (i.p. route) with 5 × 105 PFU of lacZ-tagged, mck-expressing RM427+. Mean (line) and standard deviation are indicated along with data points from individual mice. B. Titers of virus in spleen, lungs and liver from the mice used in A. C. PECs recruited by virus infection in peritoneal exudates 96 h post inoculation with 5 × 105 PFU of mck-expressing virus RM427+ (performed in parallel with A and B). The percentage of virus-positive cells in each sample is indicated in parentheses. D. Time course of infection in SGs following inoculation (FP route) of groups of three BALB/c (open triangle) or MCP-1-/- mice (open diamond) with 107 PFU of the mck-mutant virus RM461. For comparison, the day 14 titers are shown for groups of three BALB/c (filled square) or MCP-1-/- mice (filled diamond) inoculated with 107 PFU of RM427+.
