(A) U266 and NCI-H929 cells were treated with the indicated concentrations of BFA for 48h. Each bar represents the mean of 3 independent experiments, error bars indicate the SEM. (B-C) Time course of brefeldin A-induced apoptosis in U266 (B) and NCI-H929 (C) multiple myeloma cells. Cells were treated for 24 h and 48 h with the indicated concentrations of brefeldin A. Apoptosis was measured by Annexin-V-FITC/PI double-staining (shown) and active caspase-3 staining (data not shown). n =3, error bars indicate the SEM for each condition. (D) U266 and NCI-H929 myeloma cells were treated with 30 and 100 ng/mL brefeldin A for 16 h. Secreted levels of VEGF were quantitated in the culture medium by ELISA as described in the Materials and Methods. Bars represent the mean of 2 assays, error bars indicate the SEM.
