    A. Hoxa-9-/- progenitors show normal homing to the bone marrow. Wild-type and Hoxa-9-/- marrow cells were injected into lethally irradiated animals, which were sacrificed 15 hours later. Marrow cells from their femurs were analyzed for myeloid progenitors in standard clonogenic assays in semisolid media. The total number of progenitors per femur was calculated and compared to the number of progenitors initially injected to determine the percentage of progenitors that had homed to the marrow. Bars represent standard error. B. and C. Lin-sca-1+ cells from Hoxa-9-/- marrow localize to the endosteal zone. Lin-sca-1+ marrow cells were labeled ex vivo with the fluorescent dye CFSE and injected into non-irradiated mice. The recipient mice were sacrificed 15 hours later and their femurs were sectioned for examination under a fluorescent microscope to determine the localization of the transplanted cells. B. Cells within twelve layers of the endosteum scored as residing in the endosteal region, whereas cells more distant were scored as residing in the central region. C. Photomicrograph of marrow section. The white arrowheads in Panel C indicate cells located within the endosteal layer, while cells marked by white arrows are located in the central region. The black arrows indicate the endosteum, with cortical bone shown to the left. Some separation artifact has occurred with fixation and sectioning.
