Survival of wt, Apaf-1—/—, Caspase-9—/— and bcl-2—transgenic fetal liver-derived mast cells deprived of either (A) IL-3 or (A) SCF. Cell viability was measured by exclusion of PI. Graphs indicate mean +/- SD (n=19 wt, n=4 Apaf-1—/—, n=4 Caspase-9—/—, n=3 vav-bcl-2).

Analysis of Caspase-6 and Caspase-7 processing as well as ICAD cleavage in cytokine deprived mast cells from wt, Apaf-1—/—, Caspase-9—/— and vav-bcl-2 transgenic mice. Cells were either cultured in medium containing IL-3 + SCF or deprived of both cytokines for 48 h. Western blotting was used to demonstrate processing of Caspase-6 and Caspase-7, seen as disappearance of ∼p35 pro-caspase bands and in the case of Caspase-7 also as appearance of the cleaved ∼p18 fragment. Western blotting also shows cleavage of ICAD (inhibitor of he caspase-activated DNAse CAD, seen as disappearance of full-length p37 protein. Asterisks indicates non-specific band.
