Jurkat cells were treated with the indicated concentrations of LZ-CD95L or left untreated for 16 h. Apoptosis was analyzed in parallel by FSC/SSC analysis (left panels) and according to the method of Nicoletti et al. (right panels). Numbers in FACS plots represent percentage of cells in the apoptosis gate.

A) HTLV-1 infected cell lines ATL-3, CHAMP and SP, which were derived from patients, were incubated for 16 h with the indicated concentrations of agonistic anti-CD95 antibody anti-APO-1. Apoptosis was determined by analyzing subdiploid-DNA by FACS. Specific apoptosis was calculated as decribed in Material and Methods. B) Expression levels of Tax in HTLV-1 infected cells. Lysates of non-infected (CEM) and HTLV-1 infected (HuT-102, ATL-3, CHAMP, and SP) cells were subjected to SDS-PAGE and Western blot analysis with antibodies against Tax. Analysis of Erk expression served as control for equal protein loading. C) Expression of c-FLIP and Bid in HTLV-1 infected cells. Lysates of CEM, HuT-102, ATL-3, CHAMP, and SP cells were subjected to SDS-PAGE and Western blot analysis with antibodies against c-FLIP and Bid. Analysis of Erk and FADD expression served as control for equal protein loading.
