A recombinant single-chain IL-7/HGFbeta hybrid cytokine induces juxtacrine interactions of the IL-7 and HGF (c-Met) receptors and stimulates the proliferation of CFU-S12, CLPs, and pre-pro-B cells
Blood Lai et al. 107: 1776

Supplemental materials for: Lai et al

Files in this Data Supplement:

  • Figure S1. Western analysis of purified scIL-7/HGFβ (JPG, 159 KB) -

Lane 1: inactive form, rabbit anti-IL-7 antibody (from Santa Cruz Biotech); lane 2: inactive form, rabbit anti-HGF antibody (from Santa Cruz Biotech); lane 3: active form, goat anti-IL-7 antibody (from R&D systems); lane 4: active form, goat anti-HGF antibody (from Santa Cruz Biotech).

  • Figure S2. Differential stimulation of pre-pro-B, pro-B, and pre-B cells by rIL-7 and scIL-7/HGFβ (JPG, 127 KB) -

Freshly harvested rat BM cells were cultured in RPMI-1640 containing 2-ME in the presence of 10 ng/ml of rIL-7 or 30 ng/ml of rIL-7/HGF or both. Nonherent cells were harvested at day 14 and analyzed by flow cytometry. (Left) The B220+ population was subdivided into CD43+ (pre-pro-B and pro-B) and CD43 (pre-B/immature B) cell populations. (Right) The B220+ CD43+ population was further divided into HSA (pre-pro-B) and HSA+ (pro-B) cell subsets. The proportions of cells in each of these fractions are indicated.
