Clarifying the role of Stat5 in lymphoid development and Abelson-induced transformation Files in this Data Supplement: Two Stat5a/b+/+, eight Stat5a/bΔ/+ and eight Stat5a/bΔN/ΔN Ab-MuLV transformed cell lines were examined for Stat5a/b expression by Western Blot analysis. (A) Percentages of lin—/c-kit+/sca-1+ cells in Stat5a/b+/+ and Stat5a/bnull/null fetal livers. (B) Percentages of LT-HSCs (CD34—) and ST-HSCs (CD34+) gated on lin—/c-kit+/sca-1+ cells. Stat5a/bnull/+ (n=5) and Stat5a/bnull/null (n=4) fetal liver cells (ED 14) were co-cultivated on an irradiated (15Gy) MEF feeder layer in the presence of IL-7 (10 ng/ml). Generation of pro B cell stages (fraction A-C) and pre B cells (fraction D) is depicted. (A) Hematoxiline/eosine-stained histological sections of livers and spleens (magnification: 100×).
