    Figures are representative for at least 3 independent experiments consisting of 3 mice. (A) Absolute numbers of splenocytes and thymocytes from wild type (wt) and Gimap4 ko mice were counted. (B) Thymic subsets were determined by four colour flow cytometry from wt and Gimap4 ko mice and statistically analyzed. (C) Developmental T cell subsets of thymus and T/B cell ratio of spleen were determined by flow cytometry. Genotypes are displayed above the dot plots; numbers within the quadrants indicate percentages. (D) Flow cytometric analysis of thymocytes from HA TCR transgenic (tg) and HA tg/Gimap4 ko mice illustrate normal allelic exclusion. (E) Frequency of H2-Db-NP366-374 tetramer positive cells among CD8+ T cells in blood of C57BL/6 and Gimap4 ko mice at the indicated days after influenza A infection. (F) IFN production of CD8+ T cells from after 12 days of in vitro stimulation with influenza-specific peptide was determined by intracellular IFN staining of CD8+ T cells. (G) T cells of C57BL/6 and Gimap4 ko mice were sorted to high purity and activated by incubation on plate-bound anti-CD3 and soluble anti-CD28 for 3 days. Cytokine profile was determined by Cytokine Bead Array Becton Dickinson.

    Enriched T cells from Gimap ko mice (empty circles) and heterozygous littermates (filled circles) were incubated either in the absence of serum, or with the apoptotic stimuli etoposide 10 µg/ml, gamma irradiation 2 Gray and dexamethasone 1 µM. Percentage of cells belonging to live, apoptotic or dead population were measured by flow cytometry using fluorochrome-Annexin V and PI labelling. Figures present 3 independent mice and their averages (wild-type, dashed line; ko, continuous line).
