Cellular localization and trafficking of tissue factor

    Fibroblasts (WI-38) were first treated with either control buffer, methyl--cyclodextrin (CD) (10 mM) for 45 min (to deplete the membrane cholesterol and caveolae) or filipin (F) (5 µg/ml) for 15 min (to disrupt caveolae without depleting the membrane cholesterol) before they were immunostained with polyclonal anti-human TF and monoclonal human caveolin-1 antibodies. The overlap of green (caveolin-1 staining) and red (TF staining) fluorescence was used to evaluate the extent of TF co-localization in lipid rafts and caveolae. The inset shows digital magnification of a small portion of plasma membrane.
