Roles of tyrosine 589 and 591 in STAT5 activation and transformation mediated by FLT3-ITD
Blood Rocnik et al. 108: 1339

Supplemental Figures for: Rocnik et al

Files in this Data Supplement:

  • Figure S1. Representative mass spectrometry data of peptides before and after tyrosine phosphorylation (JPG, 220 KB) -
    Phosphorylation site determination for tyrosines 591 (A), 726 (B) and 955 (C) are shown.

  • Figure S2. STAT5 and FLT3 activation by FLT3-ITD Y589F (JPG, 35.7 KB) -
STAT5 (A) or FLT3 (B) activation was examined by immunoblot analysis of Ba/F3 cell lysates starved of IL-3 for 4 hours. For FL stimulation, starved cells were treated with 100ng/ml FL and incubated for 10 minutes at 37°. For stimulation with IL-3, (Ba/F3 -/+) starved cells were treated with 10ng/ml IL-3 and incubated for 10 minutes at 37°. Total cell lysates were immunoprecipitated with an either an anti-STAT5 or anti-FLT3 antibody and separated by SDS-PAGE. Immunoblot analysis was conducted with an anti-phosphotyrosine (4G10) antibody (top panels). The blots were then stripped and reprobed with anti-STAT5 or anti-FLT3 antibody (bottom panels).
