32P end-labelled oligonucleotides corresponding to a consensus pre-mRNA intronic 3′ polypyrimidine tract or to a scrambled sequence (lanes 5-8) of equivalent length and base composition were incubated with oligo alone (lane 1,5), with purified recombinant PSF alone (lane 2,6), purified recombinant ALK kinase domain alone (lane 4,8), or with PSF that had been preincubated with ALK kinase domain under conditions to allow tyrosine phosphorylation (lane 3,7). RNA-protein complexes were resolved by native-PAGE and visualized by autoradiography.
