PI3K regulates pleckstrin-2 in T-cell cytoskeletal reorganization

  • Video 1. Effect of Latrunculin-A on pleckstrin-2-induced cellular spreading (AVI, 5.32 MB) -
    Jurkat cells expressing GFP fused to the amino-terminus of wild type pleckstrin-2, were plated on OKT3. Real time confocal imaging of live cells was performed. Shown is a cell expressing pleckstrin-2, before and after inhibition of the actin cytoskeleton with Latrunculin-A for 60 seconds. Images were captured at ∼10 second intervals and are displayed at 5 frames per second. The duration of observation was ∼14.5 minutes, and Latrunculin-A was added after imagining the cells for ∼5 minutes.

  • Video 2. Effect of Wortmannin on the membrane association of pleckstrin-2 in Jurkat cells (AVI, 362 KB) -
    Jurkat cells expressing GFP fused to the amino-terminus of wild type pleckstrin-2, were plated on fibronectin. When indicated, Wortmannin was added to the chamber slide to a final concentration of 100 nM. Images were captured at ∼15 second intervals, and are displayed at 5 frames per second. The duration of observation was ∼13 minutes, and Wortmannin was added after imaging the cells for ∼4 minutes.

  • Video 3. The membrane association of pleckstrin-2 in 2B4 cells is disrupted by LY294002 (AVI, 156 KB) -
    2B4 T-cells expressing GFP fused to the amino-terminus of wild type pleckstrin-2, were plated on fibronectin. Images were captured at ∼20 second intervals, and are displayed at 5 frames per second. The duration of observation was ∼8 minutes, and 50 µM LY294002 was added after imaging the cells for ∼4 minutes.
