Human blood was perfused over a vWF/collagen coated surface at a shear rate of 1000 s-1 for 4 min. Thrombi formed on collagen, were then post-perfused for 10 min with PPACK-anticoagulated plasma, while phase contrast images were captured.

Citrated human blood was recalcified with CaCl2/MgCl2 before flowing over a vWF/collagen surface at a shear rate of 1000 s-1. Fibrin-containing thrombi formed on collagen were post-perfused for 10 min with HEPES buffer pH 7.45, containing 2 mM CaCl2 and 2 mM MgCl2, supplemented with AR-C69931MX (30 µM).
