- Figure S1. DLI do not mediate GVL effects in full chimeras when donors and recipients share Class I MHC locus (JPG, 62.1 KB)

Data were pooled from two independent experiments. B6 mice were lethally irradiated and reconstituted with B10.MBR TCD BM to generate FC. In this strain combination, both donor and recipient share Kb. On Day 56, FC received 3 × 107 B10.MBR SC and 1 × 103 EL4 cells were given one week later. FC receiving DLI (closed triangle, n = 8), EL4 (*, n = 14) or both (open box, n = 21) are shown.
- Figure S2. Accumulation of graft-versus-host-reactive 2C CD8+ T cells is dependent upon host hematopoietic APC (JPG, 46.1 KB)
-

1 × 107 B6 CD45.1 SC and 1 × 106 2C CD8+ T cells were transferred to B6 CD45.2 + BALB/c → BALB/c MC or B6 CD45.2 → BALB/c FC 10 weeks following BMT. Absolute numbers of CD8+1B2+ T cells in recipient spleens of MC and FC on Day +12 following DLI (n=9 each group).
- Figure S3. Accumulation of graft-versus-host-reactive donor CD4+ T cells is dependent upon host hematopoietic APC (JPG, 50 KB)
-

1 × 107 B6 CD45.1 SC and 1 × 106 2C CD8+ T cells were CFSE-labeled and then transferred to B6 CD45.2 + BALB/c → BALB/c MC or B6 CD45.2 → BALB/c FC 10 weeks following BMT. Absolute numbers of CD45.2-CD4+ V 3+ or CD45.2- CD4+ V 8.1/2+ T cells in recipient spleens of MC and FC on Day +12 following DLI (n=3 each group).
- Figure S4. IFN-γ expression by donor CD4+ T cells is independent of the presence of host hematopoietic APC (JPG, 97.7 KB)
-

Representative flow cytometric plots of intracellular staining of IFN- in gated CD45.2-CD4+ T cells in recipient spleens of MC and FC on Day +12 following DLI after brief ex vivo stimulation. Percentages of gated CD45.2-CD4 cells that were IFN- + were 53 ± 5% in MC versus 46 ± 3% in FC (p= NS, mean ± SEM, n=3 each group).
