Triple-color interphase FISH on a primary myeloma tumor, which has 4 copies of 1q21 (red) compared to 2 copies of 1q31 (green). White circle indicates a cIg (cytoplasmic immunoglobulin) negative cell’s nucleus, which shows a normal pattern of FISH signals (Figure S1A). Metaphase FISH analyses were performed on human myeloma cell lines. Delta47 shows tandem duplications of 1q21 (Figure S1B), while RPMI8226 does not (Figure S1C).

In patients with ISS stage I (left panel) and ISS stage II (middle panel), those with Amp1q21 had inferior OS compared with those lacking Amp1q21. However, in patients with ISS stage III (right panel), OS was similar between those with Amp1q21 and lacking Amp1q21.

This Excel workbook contains five separate worksheets: (A) contains data on 14 MGUS cases, (B) contains data on 31 SMM cases, (C) contains data on 479 newly diagnosed MM, (D) contains data on 31 relapsed MM treated on Total Therapy, and (E) contains data on 28 paired cases from diagnostic/pretreatment samples and paired relapsed MM treated on Total Therapy 2. The first column of each worksheet contains the sample id representing a unique sample identifier and each row a sample. The second, third, fourth, fifth, and sixth columns indicate the percentage of clonal plasma cells in each sample with the indicated copy numbers of 1q21. The seventh column indicates the percentage of cells within each sample with greater than 2 copies of the 1q21 locus. The eighth column indicates the final call of number of copies within each sample based on method described. The ninth column in worksheet (A) and (B) indicate the cases that progressed to MM within the study period. In worksheet (E) there are two tables; the left represents the diagnostic/pretreatment samples and the right the relapsed samples from the same patients.
