    HEK-293 cells were UV irradiated (100 mJ) and analyzed in a flow cytometer for induction of apoptosis after overnight incubation. (A) Unstained control. (B) Annexin V/propidium iodide (PI) staining. The percentage of PI-positive cells is shown in the upper right corner.

  • Figure S2. Phagocytosis of apoptotic HEK-293 cells by DCs (PDF, 114 KB) -
    Uptake of apoptotic HEK-293 cells by DCs was analyzed in phagocytosis experiments. Membrane-dye–labeled apoptotic HEK-293 cells (red) (A) and immature DCs (green) (B) were cocultured for 24 hours and analyzed in FACS. The double-positive fraction resembles DCs that have taken up apoptotic cells (D). As control, phagocytosis was performed at 4°C (E) and, in addition, labeled cells were mixed shortly before FACS analysis (C) to exclude unspecific diffusion of membrane dyes. The data shown are representative of 5 independent experiments.

  • Figure S3. Uptake of apoptotic material by DCs (PDF, 34.7 KB) -
    Confocal laser scanning microscopy was performed to analyze the uptake of apoptotic HEK-293 cells (red) by DCs (green). Phagocytosis of cellular material is indicated by the colocalization (yellow) of the PKH membrane dyes.
