Differential effects of recombinant thrombopoietin and bone marrow stromal-conditioned media on neonatal versus adult megakaryocytes

    Concentrations of 79 cytokines were measured in stromal-conditioned media (CM) using a cytokine antibody array system (RayBio Human Cytokine Antibody Array, V; RayBiotech, Norcross, GA) according to the manufacturer’s instructions. The signal intensity of each individual spot in the antibody array membrane was determined by densitometry using Scion Image software (Scion, Frederick, MD), and was then normalized against the intensity of the positive control signal in each membrane. For each CM sample, the corresponding UCM was also assayed, and its values were subtracted from the CM. Results presented in the table represent the means and standard deviations of 4 separate experiments and are listed in order of decreasing concentrations.
