Stably transfected CHO cells expressing CD32A-R131 (left panels) or CD32B (right panels) were incubated in PBS-1%BSA-SA with 3ug/ml of aglycosylated, chimeric 2B6 (ch2B6-N297Q), aglycosylated, humanized 2B6 (hu2B6-3.5-N297Q), aglycosylated IV.3 (IV.3-N297Q) or FLI8.26 (bold lines). Isotype control antibody staining is represented by the dotted lines. Primary antibodies were washed twice in PBS-1%BSA-SA and bound antibody was detected using PE-labeled goat F(ab′)2 anti-mouse (H+L) IgG or PE-labeled goat F(ab′)2anti-human Fc-specific secondary antibody. Samples were then washed with an excess of PBS and then analyzed by flow cytometry using a FACS Calibur.
