B. >2.5× in iDCs + IFN iDC + IFN signal value >100, present; <1.5× in mDCs + IFN .

- Figure S1. IFNα/β early alters CD40L gene induction profile (PDF, 14.5 KB) -
Affymetrix data was queried for genes induced by CD40L that were inhibited by exposure of iDCs to IFN / during maturation. The top nine differentially regulated genes are displayed graphically by relative expression in iDCs, mDC, or mDCs exposed to CD40L (mDC + 40L). Blue line, untreated. Red line, IFN / treated.
- Figure S2. IFNα/β does not alter CD40L induced IκB-α phosphorylation or degradation (PDF, 24.9 KB) -
iDCs matured in the presence of 60 IU/ml IFN / , mDCs, and mDCs exposed to 60 IU/ml IFN / were incubated with 0.5µg/mL CD40L for 30 minutes. 20µg of protein from the total cell lysates were run by 12% SDS-PAGE, transferred to PVDF membranes, and blotted with indicated antibodies as indicated in “Materials and Methods.”
