(A) Adult and cord blood moDCs (106 cells) were collected and directly lysed in Laemmli sample buffer. Equal volumes of total cell lysate were resolved by 8% SDS-PAGE and analyzed by Western blotting using antibodies directed against MyD88 (F-19; Santa Cruz Biotechnology), TRIF (ALX-210-908; Alexis Biochemicals, Zandhoven, Belgium), TRAM (ALX-210-914; Alexis Biochemicals), IRF-3 (550428; BD Biosciences, Erembodegem, Belgium), TBK1 (IMG-139; Imgenex, Huissen, The Netherlands), and IKK (IMG-270A; Imgenex). To control loading, immunoblots were reprobed with anti-GAPDH (Biodesign International, Venendaal, The Netherlands). Data are representative of 12 different donors in each group. (B) TBK-1 and IKK mRNA levels were determined in adult and cord blood moDCs by real-time RT-PCR.
