Hypoxia-inducible transcription factor–1 alpha determines sensitivity of endothelial cells to the proteosome inhibitor bortezomib

    HUVEC were cultured either in normoxic or hypoxic (5% O2) conditions for 1 day, and where indicated, MG132 (7.5 mM) or bortezomib (100 nM) was added for the last 12 hours. The percentage of living cells, calculated as annexin V/PI double-negative, was assessed by FACS analysis. The MG132/bortezomib-induced apoptosis was greater when HUVEC were cultured in hypoxia compared with the normoxic/untreated counterpart. Data represent the mean plus or minus SEM of 3 independent experiments, and statistical analysis was performed with the Wilcoxon/Kruskal-Wallis test. **P < 0.5.
