(B) In vitro–translated Gfi1b was incubated with each of the indicated double-stranded oligos (200× or 1000× excess) for 15 minutes prior to addition of radiolabeled consensus Gfi1b-binding oligo. Only the consensus Gfi1 oligo (200× excess) competed with the consensus oligo for binding to Gfi1b. In direct EMSA experiments Gfi1b showed detectable binding to the consensus oligo but not to Gfi1bBS-1, BS-2, or BS-3.
