Control and GILZ-transfected DCs were pulsed with PPD and used to stimulate CD4+ T lymphocytes either unfractionated or depleted of CD45RO+, CD45RA+, or CD25+ cells. (a) Expression of IL-10 by CFSElo CD4+ T lymphocytes after stimulation with control (ν) or GILZ-transfected (-) DCs in a representative of 3 experiments. Control mAb staining obtained with GILZ-transfected DCs is shown as a dotted line. (b) CD4+ T lymphocytes were recovered at the end of the stimulation and added to CFSE-labeled PBMCs stimulated with PPD. Proliferation of CD4+ responders was analyzed in 3 independent experiments.

(a) Increasing numbers of CD25+CD4+ T lymphocytes were added to CD25–CD4+ T lymphocytes and to PPD-pulsed DCs transfected with either the control (□) or the GILZ-encoding (ν) vector. The fraction of IL-10–expressing CD4+ T lymphocytes was determined on day 7. Results (mean ± SEM) are from 3 experiments. (b) Purified CD4+CD25+ T lymphocytes were stimulated with PPD-pulsed DCs transfected with either the control (ν) or the GILZ-encoding (-) vector. IL-10 expression by CD4+ T lymphocytes was determined 7 days later.
