Isolation of resident monocytes and inflammatory monocytes by magnetic cell sorting (JPG, 89.7 KB) - Resident monocytes (CD16+CD14low) were enriched from PBMCs by negative selection of T, B, and NK cells (using a cocktail of CD3, CD19 and CD56 MicroBeads), followed by positive selection by CD16 MicroBeads. Inflammatory monocytes (CD16–CD14+) were enriched by negative selection of CD16+ cells, followed by positive selection by CD14 MicroBeads. Cells are shown after purification. Note that the CD16+CD14low resident monocytes were highly enriched in TIE2+ cells, whereas the CD16–CD14+ inflammatory monocytes contained few TIE2+ cells. Figure S2. Circulating TIE2+ monocytes have marked pro-angiogenic activity in vivo (JPG, 35.7 KB) - Human glioma U87 cells were injected subcutaneously into nude mice with or without the indicated monocyte populations at 1:100 or 1:20 ratio. Tumours were grown for 5-7 days and analyzed by CD31 immunostaining and confocal analysis to assess angiogenesis. Representative tumour images reconstructed by superimposition of overlapping confocal planes acquired at 100× magnification. Scale bar as indicated.
