Crosstalk between RANKL and Fas signaling in dendritic cells controls immune tolerance
Blood Izawa et al. 110: 242

Supplemental materials for: Izawa et al, Volume 110, Issue 1, 242-250

Files in this Data Supplement:

  • Figure S1. Effect of RANK siRNA on Fas expression and apoptosis through RANKL and Fas in DCs (PDF, 263 KB) -
    (A) Experimental protocol of RANK siRNA transfection using BMDCs from MRL+/+ and MRL/lpr mice is shown. (B) RANK expression on BM cells and RANK siRNA-transfected BMDCs from MRL+/+ and MRL/lpr mice was analyzed by flow cytometric analysis. Irrelevant oligonucleotides were used as control. Data are representative of 3 independent experiments. (C) RANK expression of RANK siRNA-transfected BMDCs from MRL+/+ and MRL/lpr mice was detected by immunoblot. Irrelevant oligonucleotides were used as control. GAPDH was used as a control for loading. Results are representative of 3 independent experiments. (D) Fas expression on RANK siRNA-transfected BMDCs from MRL+/+ and MRL/lpr mice was analyzed after the stimulation of RANKL. Irrelevant oligonucleotides were used as control. Numbers above horizontal lines indicate percentage of Fas+ cells. Data are representative of 3 independent experiments normalized to the control values. (E) Inhibitory effect of RANK siRNA on RANKL-induced apoptosis of BMDCs from MRL+/+ and MRL/lpr mice was observed. Apoptotic cells were detected by flow cytometric analysis. Numbers above horizontal lines indicate percentage of annexin-V+ cells. Results are representative of 3 independent experiments.
