Cells from lymph nodes (LNs) and spleens were prepared from naive B6 mice, and were stained with FITC-labeled mAb to CD11c (clone HL3) or CD11b (clone Mac1), PE-labeled mAb to murine B7-H1 (clone MIH5), or APC-labeled mAb to CD3 (clone 2C11) as indicated. The samples were analyzed by flow cytometry. The data are from 1 of 3 representative experiments.

To test specificity of B7-H1 mAb (clone 10B5), CHO cells expressing mouse PD-1 (mPD-1/CHO) were incubated with B7-H1Ig fusion protein premixed with anti–B7-H1 (10B5) mAb (red line) or hamster Ig (green line). Filled line indicates negative control. Cells were subjected to flow cytometry analysis and the results are expressed in the histogram (top panel). To test specificity of B7-DC mAb (clone YL-1), mPD-1/CHO cells were incubated with B7-DCIg fusion protein premixed with anti–B7-DC (YL-1) mAb (red line) or rat IgG (green line), and were analyzed as described above. Filled line indicates negative control.
