    (A) The 5′ untranslated region (UTR) encoded by the Gata2 1G exon contains 2 out of frame AUG codons embedded in consensus translation initiation motifs.55 Translation of upstream open reading frames 1 and 2 (uORF1 and uORF2) would result in short peptides of 22 and 63 amino acids (aa), respectively. uORF2 overlaps the translation initiation codon of Gata2 (Gata2 ORF). (B) Putative secondary structure of the Gata2 1G 5′UTR calculated by the Mfold computer program.57 The 1G 5′UTR may adopt a Y-shaped secondary structure embedding the AUG codons of the uORFs and the canonical Gata2 start codon. The two most prominent loops in this structure are potential binding sites for poly(rC) binding protein 1 (PCBP1) and polypyrimidine tract binding protein (PTB), proteins known to be involved in the control of translation initiation.56 Red bars: GC base pairs; blue bars: AU/GU/AC base pairs; numbers indicate nucleotide positions.
