Detection of human IL2RG in bone marrow from SCID-X1 mice transduced with the A2UCOE-IL2RG lentiviral vector (PDF, 30.6 KB) - Total protein extracted from bone marrow of SCID-X1 mice transduced with the A2UCOE-IL2RG was subjected to Western blotting and analyzed using antibodies to either human IL2RG (R&D System, Abingdon, UK) or mouse actin, which acted as a loading control for each sample. Bound antibodies were detected using the UVICHEMI imager system (Uvitech, UK). Lane 1: untransduced SCID-X1 mouse; Lanes 2-6: A2UCOE-IL2RG vector transduced SCID-X1 mice; Lane 7: human ED7R cells deficient in IL2RG; Lane 8: human ED7R cells stably expressing IL2RG as a positive control. Note that the human IL2RG protein was detected in all SCID-X1 mice transduced with the A2UCOE-IL2RG vector.
