(A) Kinetic pattern of cyclin expression during the cell cycle. Cyclin E1 is expressed in mid-to late G1, with a characteristic Gaussian profile. After Cyclin E degradation, Cyclin A synthesis occurs, with an exponential behavior. mRNA levels for (B) Cyclin E1 and (C) Cyclin A2, in BL cell lines, either transfected with RBL2/p130 or the empty vector. Cells were harvested 48 hours and 72 hours (bars 2, 3) after transfection and compared to the time 0 (bar 1). Please note that in RBL2/p130-transfected cells, the expression kinetic of Cyclin E1 and Cyclin A2 resembled the physiological pattern of late G1 phase, whereas control cells presented with patterns characteristic of S phase (B,C). (D) mRNA levels for E2F-4 in BL cells, either transfected with RBL2/p130 or the empty vector. Cells were harvested at 24, 48, and 72 hours (bars 1, 2, 3) from transfection.
