Cleavage of tetrapeptide substrates does not distinguish between individual caspases (JPG, 58.3 KB) - The indicated units of recombinant caspase-3 (A) and caspase–10 (B) were incubated with 50 µM ac-AEVD-afc (upper panels) or ac-IETD-amc (lower panels). 1 unit of recombinant caspase catalyzes the cleavage of 1 nmol IETD-substrate at 37°C per hour. The specific activities of caspase-10 and caspase-3 were 3300 and 277000 units per mg protein, respectively. Substrate cleavage was monitored for up to 3 h. The results show that the alleged caspase-10-specific substrate ac-AEVD-afc is more efficiently cleaved by caspase-3 than by caspase-10.
